"Guidelines for indoor bioassay tests of pesticides" is a series of standards. This part is Part 6 of the Guidelines for Indoor Bioassay Tests of Pesticides - Insecticides.
This standard is proposed and managed by the Ministry of Agriculture of the People's Republic of China. The drafting unit of this standard is the Pesticide Control Institute of the Ministry of Agriculture. The main drafters of this standard are Wang Xiaojun, Ou Xiaoming, Lin Ronghua, Ni Yaping, Jiang Hui, and Zhang Hong. 1 Scope
Guidelines for Indoor Bioassay Tests of Pesticides - Insecticides Part 6 Insecticidal Activity Test - Insect Immersion Method
This part specifies the basic requirements and methods for insecticide activity tests using the insect immersion method.NY/ T1154.6—2006
This part is applicable to the indoor test for pesticide registration for the determination of insecticide activity. Other tests shall be carried out in accordance with this part. 2 Test conditions
2.1 Test targets
Test targets shall be selected from Lepidoptera larvae of the same age and sensitivity, such as Mythimnasebarata, Chilosuppres-salis, Plutella xylella, etc., Homoptera insects such as aphids, leafhoppers, planthoppers, etc., Coleoptera adults such as potato moths (Henosepilachna vigintioctomaculata), Thysanoptera such as thrips, etc. 2.2 Instruments and equipment
2.2.1 Electronic balance (sensitivity 0.1mg); 2.2.2 Immersing insect cage;
2.2.3 Volumetric flask;
2.2.4 Culture blood with a diameter of 9cm;

3 Experimental design
3.1 Test material preparation
Select sensitive test insects that are raised indoors and have the same age, such as 2nd or 3rd instar larvae for Lepidoptera, nymphs or wingless adults for Homoptera, and young adults for Coleoptera. 3.2 Reagents||tt ||3.2.1 Test Agent
The test agent uses the original drug (mother drug) and indicates the generic name, trade name or code, content, and manufacturer. 3.2.2 Control Agent
Control agents use the original drug that has been registered and commonly used in production. The chemical structure type or mode of action of the control agent should be the same or similar to that of the test agent.
3.3 Test Procedure
3.3.1 Agent Preparation
Water-soluble agents are directly dissolved and diluted with water. Other agents are dissolved with appropriate organic solvents (such as acetone, chloroform, ethanol, etc.) and then diluted with 1

water solution containing an appropriate amount of surfactant such as 0.1% Tween 80 (or 0.1% Triton-X100). According to the activity of the agent, set 5 to 7 series of mass concentrations according to the method of geometric ratio or arithmetic difference. The amount of liquid medicine for each mass concentration shall not be less than 50mL. 3.3.2 Treatment with pesticides
After immersing the target insects in the liquid medicine for 5s~10s, absorb the excess liquid medicine with filter paper and transfer the test insects to normal conditions for breeding. Each treatment is repeated 4 times, and 10~20 insects are immersed in each repetition. A treatment with a corresponding organic solvent without pesticide is set as a control. 4 Investigation
Investigate the mortality of the test insects 24h after treatment, and record the total number of insects and the number of dead insects. The investigation time can be shortened or extended according to the test requirements and the characteristics of the pesticide.
5 Data statistics and analysis
5.1 Calculation method
Based on the survey data, calculate the corrected mortality rate of each treatment in percentage (%). Calculate according to formula (1) and (2), and keep the results to two decimal places:

If the control mortality rate is <5%, no correction is required: if the control mortality rate is between 5% and 20%, correction should be made according to formula (2): if the control mortality rate is >20%, the test needs to be repeated.
5.2 Statistical analysis
Use software such as statistical analysis system (SAS) and data processing system (DPS) to analyze and calculate the LCs0, LCou, b value (standard error) and 95% confidence limits of each agent to evaluate the stomach poison activity of the test agent against target insects. 6 Results
Analyze and evaluate the statistical results, write a formal test report, and list the original data. 2
