Guidelines for Testing Specificity, Consistency and Stability of New Plant Varieties Persimmon
This standard specifies the technical requirements for testing specificity, consistency and stability of new persimmon varieties and the general principles for judging the results. This standard applies to the testing of new varieties of persimmon (Dio. spyroskaki Thunb.). 2 Normative References
The following documents are indispensable for the application of this document. For all referenced documents with dates, only the versions with the dates are applicable to this document. For any undated referenced document, its latest version (including all amendments) shall apply to this document. The terms and definitions defined in GB/T 19557.-1 and the following terms and definitions apply to this document. 3.1
Individual measurement.- mneasorement of a nurnber of individual plants or parts of plants. A group of individual records are obtained by measuring a certain organ or part of a batch of plants. 3.2
Group visual inspection: tissment by a single chservation of a group of plants or parts of plants. A group record is obtained by visually inspecting a certain organ or part of a batch of plants. 4 Symbols
The following symbols apply to this document,
MS: individual measurement
VG: population measurement
QL: quality trait, bzxz.net
QN: quantitative trait,
PQ: pseudo-quality trait.
*: The trait marked as P()V is used for the important traits required for variety description. Unless the expression state of the trait cannot be tested due to environmental conditions, all P()V members should use these traits. (a)-~(d): The marked content is explained in detail in B.2 (i); The marked content is explained in detail in B.3: The underline in this document is a special reminder of the scope of application of the test trait. 5 Requirements for propagation materials
5.1 Propagation materials are provided in the form of seedlings or scions. 5.2 The number of seedlings or scions submitted shall be at least 10 plants (branches). 5.3 The submitted propagation materials should be healthy in appearance, high in vitality, and free from pests and diseases. Seedlings should be one-year-old grafted seedlings, and the species of brick and wood should be marked. Grafting NY/T 2522—2013
Secrets should be healthy dormant vegetative scions, and each scion should have at least IC effective buds. 5.4 The propagation materials submitted are generally not subjected to any treatment that affects the expression of variety traits. If they have been treated, detailed descriptions of the treatment should be provided.
5.5 The propagation materials submitted should comply with the relevant provisions of China's plant quarantine. 6 Test methods
6.1 Test cycle
The test cycle is at least 2 independent annual growth cycles. Normal fruits should be produced before the end of each growth cycle. A complete growth cycle of persimmon refers to the entire growing season from the beginning of budding (flower buds and/or vegetative buds), through flowering, fruit ripening, entering dormancy, and until the end of the dormancy period (new buds swelling). 6.2 Test location
The test is usually carried out at one location. If some traits cannot be fully expressed at this location, they can be observed at other qualified locations.
6.3 Field test
6.3.1 Test design
Please plant the varieties and similar varieties adjacently. Seedlings are directly planted: single plant plot, 5 times of replication, row spacing 5m, plant spacing 2m. The scion is grafted onto the perennial cow's junqianzi (Dias yrosiotus L.) or other suitable rootstocks by branch grafting. 6.3.2 Field management
Carry out the normal production management method of the persimmon orchard. 6.4 Character observation
6. 4. 1 Observation period
Character observation should be carried out according to the growth and development stages listed in Appendix A. Appendix B explains these growth and development stages. 6.4.2 Observation methods
Observation of traits should be carried out in accordance with the observation methods (VG, MS) specified in Appendix A. 6.4.3 Number of observations
Unless otherwise specified, the number of samples for individual observation traits (MS) shall not be less than 5. When observing the organs or parts of the plants, the number of samples for each plant shall be 2. The entire plot shall be measured for group observation traits (VG). 6.5 Additional tests
If necessary, additional tests may be performed on traits not listed in Appendix A. 7 Determination of specificity, consistency and stability results 7.1 General principles
Specificity, consistency and stability shall be determined in accordance with the principles determined in Membrane CB/Work 157.1. 7.2 Determination of specificity
The variety under application should be clearly distinguishable from all known varieties. In the test, when the application variety has a significant and reproducible difference with similar varieties in at least one trait, the application variety can be determined to have specificity. 7.3 Determination of consistency
Use a 1% population standard and at least 95% acceptance probability. When the sample size is 5 plants, no heteromorphic plants are allowed. 7.4 Determination of stability
If a variety has consistency, it can be considered stable. Generally, stability is not tested. 2
NY/T 2522—2013
If necessary, the seedlings or scions of the variety can be replanted or grafted. Compared with the previously provided propagation materials, if there is no significant change in the expression of the trait, the variety can be determined to have stability. 8 Trait table
According to the test needs, the traits are divided into basic traits and test traits. Basic traits are traits that must be used in the test. Appendix A lists the basic traits of persimmon.
8.1 Overview
The trait table lists the trait name, expression type, expression state and corresponding code and standard variety, observation period and method, etc. 8.2 Expression type
According to the expression method of the trait, the traits are divided into three types: quality traits, pseudo-quality traits and quantitative traits. 8.3 Expression state and corresponding code
8.3.1 Each trait is divided into a series of expression states. In order to facilitate the definition of traits and standardized descriptions, each expression state is assigned a corresponding digital code to facilitate data recording, processing and the establishment and communication of variety descriptions. 8.3.2 For quality traits and pseudo-quality traits, all expression states should be listed in the test guide. For quantitative traits, in order to shorten the length of the trait table, the expression states of even codes can be omitted. The expression states of even codes can be described in the form of the previous expression state to the next expression state.
8.4 Standard varieties
The standard varieties corresponding to the expression states of some traits are listed in the trait table to help determine the different expression states of related traits and correct the differences caused by years and locations.
9 Explanation of trait table
Appendix B provides additional explanations on the observation period and observation methods of some traits in the trait table. 10 Grouped traits
In this document, the cultivar grouping traits are as follows: a) *Fruit: Fruit size (Table A.1 Neutral trait 26). b) *Fruit: Fruit shape (Table A.1 Neutral trait 27). c) *Fruit: Peel color (Table A.1 Neutral trait 28). d) *Sweet persimmon, fruit: Fruit edible maturity period (Table A.1 Neutral trait 45). e) *Astringent persimmon: Fruit: Fruit edible maturity period (Table A.1 Neutral trait 16). Fruit: Variety type (Table A.1 Neutral trait 47). f
