Guidelines for testing specificity, consistency and stability of new plant varieties This standard specifies the technical requirements for testing specificity, consistency and stability of new plant varieties, the criteria for determining test results, and the content and format of technical reports.
This standard applies to the testing and evaluation of specificity, consistency and stability of Prurus 5sPp. varieties. 2 Normative references
The clauses in the following documents become clauses of this standard through reference in this standard. For any dated referenced document, all subsequent amendments (excluding errata) or revisions are not applicable to this standard. However, parties to an agreement based on this standard are encouraged to study whether to use the latest versions of these documents. For any undated referenced document, the latest version shall apply to this standard. Terms and definitions established in GB/T19557.1~-2004 apply to this standard. 4 Requirements for materials for test varieties
4.1 Quality and quantity of test materials
Test materials are divided into scions and seedlings. The types of materials required for the applicant to submit for testing shall be determined by the approval authority based on the test requirements. If the new variety applied for testing has special uses, the quality and quantity of the test materials submitted can be determined by the approval authority based on its characteristics and scope of application.
4. 1. Scion
Scion should be healthy, free of pests and diseases, and free of quarantine objects, and obtained by non-in vitro propagation methods. The number shall be at least 50, and each root shall have at least 10 full and plump buds (excluding hidden buds). 4.1.2 Yellowwood
Seedlings shall be free of diseases and insects, quarantine objects, and healthy, with a seedling height of 0.9m~1m and a base diameter of more than 1cm. The number shall be no less than 20.
4.2 Instructions for the treatment of test variety materials
The submitted test materials shall not be treated in any way that may affect their growth without the approval of the approval authority. 4.3 Preservation of test varieties
After receiving the test materials, the testing unit shall condense them immediately for review and examination. 4.4 Others
In addition to submitting the test materials, the applicant shall also fill in the technical volume according to the appendix. 5 Test
5.1 Test time
The test duration shall be at least two normal growth cycles, and these two growth cycles shall be able to show the normal characteristics of this variety. 5.2 Test location
The conditions at the test point should be able to meet the normal growth of the plant and the normal expression of its traits. Generally, each tested variety is arranged to be tested at the same test point. If there are special requirements, multi-point testing can be carried out. For special purposes, additional tests can be set up. Additional tests should be carried out in the identification unit or specific ecological zone designated by the approval authority. 5.3 Requirements for sticky wood
The high-grafted mother trees used as rootstocks should be of the same variety, strong and healthy, free of pests and diseases, and have basically the same tree vigor, age and growth conditions. 5.4 Field design
The field design of the applied variety and similar varieties should be scientific, correct, reasonable, consistent, and have the same test conditions. The test can adopt the comparison method, inter-comparison method and high-grafting comparison method, with a plant-row spacing of 3m\u00d74m or 3m\u00d75m. Each plot should have no less than 5 plants, at least three replicates, and a protective row. In special cases, single-plant plots can also be used, with multiple replicates. 5.5 Field management
Field management measures for the test variety and similar varieties should be strictly consistent, and detailed implementation rules should be formulated. If the test is conducted in a greenhouse environment, detailed instructions should be provided.
6 Observation and discrimination of traits
6.1 Basic requirements for observation and discrimination
The approval authority shall determine the test traits of the variety based on the new variety information provided by the applicant. Variety observation shall be carried out in accordance with the requirements of Appendix A and Appendix B. The standard variety observed together with the test variety shall perform normally, otherwise, this test is invalid.
Observation records shall be carried out in accordance with the requirements of GB/T19557.1-2004. The discrimination of specificity, consistency and stability shall be carried out in accordance with the requirements of GB/T19557.1-2004. 6.2 Discrimination of specificity
If the same trait of the test variety and similar varieties is in the same code, it means that the test variety has no difference with the similar varieties in this trait, otherwise, there is a difference. If the test variety has two or more individual or quantitative traits that are different from similar varieties, or one of the quantitative traits differs from similar varieties by two or more codes, the test variety can be determined to be specific to the similar varieties. 6. 3 Discrimination of consistency
For the observation of variety consistency, the code is used as the analysis unit. If there are differences in the observed traits, the value of the trait with the most expression shall prevail and be reported. Traits that cannot be tested individually will not be subjected to consistency determination. 6. 4 Discrimination of stability
For the observation of variety stability, in the test results of the same growing season, if the expression of the same trait of the test variety is within the same code, it means that the variety is stable in this trait; otherwise, it is unstable. 7 Traits
7.1 Overview
The test traits include plant morphological characteristics, agronomic traits, quality traits and pest resistance traits, totaling 67 (see Appendix A for description of traits), divided into mandatory traits and supplementary traits. Mandatory traits are basic traits that must be observed and described for each test variety, while supplementary traits are traits that are further tested when mandatory traits cannot distinguish the test variety from similar varieties. 7.2 Mandatory traits (28)
