4 Main Reagents and Instruments
4.1 Reagents:
41-1 FLISA antigen
4.1.1-1 Specific antigen
Electrode the R3 antigen
Reo3 antigen in the serum of rats, hamsters, and hamsters. When the cells reach 10-10 years old, harvest the culture, freeze-thaw three times or sonicate. Remove the cell debris by high-speed centrifugation, and then make ELISA antibody by ultracentrifugation. 4.1.1.2 Normal antigen
BSC-1 or BHK21 tissues are frozen and thawed, and the supernatant obtained by low-speed high-speed centrifugation is sliced. 4-1-2 Antigen slice
Reo3 antigen slice BHK21 cells are cultured for 4-5 days. When the cells reach +10 years old, digest with enol, wash with PBS, and wash the slices. Dry them in room temperature and irradiate them at 20cm under ultraviolet light for 30 minutes. Cold two ketones fixed for 10 minutes, -20 ℃ storage 4.1.3 Positive serum
Reo3 original immune clean or SPF mouse, rat, guinea pig, hamster obtained antiserum, 4.1.4 Lang serum
SPF mouse, rat, urea rat, hamster serum. The General Administration of Quality Supervision, Inspection and Quarantine of the People's Republic of China approved on August 29, 2001 and implemented on May 1, 2002
4.1.5 Enzyme conjugate
GB/T14926-252001
Horseradish peroxidase alcohol labeled sheep or rabbit anti-mouse, rat, glandular greasy, hamster IgG antibody, used to detect the corresponding animal serum antibodies. Horseradish peroxidase labeled Staphylococcus aureus A (SPA) can be used for the detection of mouse, guinea pig, hamster serum antibodies. 4.1.6 Fluorescein isothiocyanate-labeled sheep or rabbit anti-mouse, rat, mouse, hamster G antibody, used to detect the corresponding animal serum antibodies, 4.2 Equipment
4.2.1 Label collection.
4.2.2 Diamond star microscope.
Ordinary foot microscope
4.2.437C wall culture box or water seeding box,
Detection method
5-1 Serological detection is carried out by ELISA method (see GB/T14926-50-2001). 5.2 Serological detection is carried out by IFA method (see GB/T14926.52-2001). 53. Use the A-shou method (see CB/19869001 for further examination) to determine the results. For positive test results, use the same method or a different method to retry. If it is still positive, it is determined to be positive. Report on the results of the determination. Make a report based on the determination results.
