Method for determining the number of Beauveria bassiana spores in workshop air (supplement)
After sampling and weighing with a dust filter, dissolve the filter with butyl acetate to form a suspension of Beauveria bassiana spores, count them under a microscope with a blood cell counting plate, and then convert them into the concentration of Beauveria bassiana spores in the air (number of spores/m\). A2 Equipment and reagents
Dust sampler; dust filter; analytical balance with a sensitivity of not less than 0.1mg, microscope; blood cell counting plate; glass rod; small beaker; butyl acetate.
A3 Procedure for determining the number of Beauveria bassiana spores
A3.1 Sampling: Use a dust sampler equipped with a dust filter to extract air at a flow rate of 1020L/min. A3.2 Preparation of bacterial solution: Place the filter membrane for collecting Beauveria bassiana spores in a small beaker, add 1-2mL of butyl acetate with a pipette, and stir thoroughly with a glass rod to make a uniform suspension of Beauveria bassiana. Then dilute it in proportion to the number of milligrams of weight gain of the filter membrane to form a mg/mL dilution. A3.3 Filling the counting pool: Place the blood cell counting plate flat on the table, place the cover glass close to the scale of the blood cell counting plate, dip the bacterial solution with a glass rod, and let it penetrate into the counting plate grid from the grooves on the front and back edges of the cover glass, absorb the bacterial solution in the grooves with filter paper, let it stand for a while, and count under a microscope. If bubbles form on the blood cell counting plate, the pool should be refilled. A3.4 Microscope counting: Counting can be performed using a 600x microscope. Count the spores in the five middle grids of a 25mm×16mm blood cell counting plate, that is, the number of spores in the four corners and the middle grid (a total of 80 small grids). For spores pressed on the double lines at the edge of the grid, count the top but not the bottom, count the left but not the right. To obtain an accurate spore count, the average of the five middle grids under the microscope should not exceed 100 at the highest and should not be less than 10 at the lowest.

Example: When the sampling flow rate is 20L/min and the sampling time is 5min, the filter membrane weight gain is 3.7mg, dissolved in 18.5ml solvent. If the average number of spores in the 5 middle grids of the counting plate is 83.3, try to calculate the number of spores/m. Substitute the above data into the formula:
