4.1 Determination of glyphosate content
4.1.1 Summary of method
After pretreatment, the sample reacts with sodium nitrite in an acidic medium to generate a glyphosate nitroso derivative, which has a maximum absorption peak at 243nm. Approved by the Ministry of Chemical Industry of the People's Republic of China on November 18, 1991 and implemented on May 1, 1992
, and can be quantified by measuring the absorbance. 4.1.2 Reagents and solutions
HG2162-91
When not specified, all reagents used are analytically pure reagents, and all water is distilled water or water of corresponding purity. 4.1.2.1 Hydrochloric acid;
4.1.2.2 Sulfuric acid solution: 50% (V/V);
4.1.2.3 Nitric acid solution: 50% (V/V);
4.1.2.4 Potassium bromide solution: 250g/L;
4.1.2.5 Sodium nitrite solution: weigh about 0.28g sodium nitrite (accurate to 0.001g), dissolve in 20mL water, the solution should be prepared before use,
4.1.2.6 Glyphosate standard sample: >99.8%. 4.1.3 Instruments
4.1.3.1 UV spectrophotometer,
4.1.3.2 Quartz colorimetric III: 1cm,
4.1.3.3 Graduated pipette: 1mL, 2mL, 5mL. 4.1.4 Analysis steps
4.1.4.1 Drawing of standard curve
4.1.4.1.1 Preparation of standard solution
Weigh about 0.30g of glyphosate standard sample (accurate to 0.0002g), transfer to a 200mL beaker, add 60mL of water, 1mL of hydrochloric acid, slowly heat to dissolve, cool to room temperature, quantitatively transfer to a 250mL volumetric flask, dilute to scale, and shake well. This solution should not be used for more than 20 days.
4.1.4.1.2 Nitrosylation
Accurately pipette 0.8, 1.1, 1.4, 1.7, and 2.0mL of glyphosate standard solution into 5 100mL volumetric flasks, and take another 100mL volumetric flask for reagent blank test.
Add 5mL of distilled water, 0.5mL of sulfuric acid solution, 0.1mL of potassium bromide solution, and 0.5mL of sodium nitrite solution to the above volumetric flasks respectively. After adding sodium nitrite, the stopper should be immediately tightened, shaken thoroughly, and left for 20 minutes (the temperature during the reaction should not be lower than 15℃). Dilute to the scale with water, shake well, and finally open the stopper and leave for 15 minutes. 4.1.4.1.3 Spectrophotometric determination
Turn on the power of the ultraviolet spectrophotometer, turn on the fluorine lamp to preheat for 20 minutes, adjust the wavelength to 243nm, use the reagent blank as a reference, and use a quartz colorimetric dish to measure the absorbance.
4.1.4.1.4 Draw a standard curve
Use the absorbance as the vertical coordinate and the volume of the corresponding standard solution as the horizontal coordinate to make sure that each point is connected into a straight line. 4.1.4.2 Analysis of glyphosate soluble powder Weigh about 0.4g of sample (accurate to 0.0002g), transfer to a 200mL beaker, add 60mL of water and 1mL of hydrochloric acid, slowly heat to keep slightly boiling for 5min, filter with fast filter paper, rinse the filter paper carefully, transfer the filtrate to a 250mL volumetric flask, cool to room temperature, dilute to scale, and shake well.
Accurately pipette 2.0mL of sample solution into two 100mL volumetric flasks, dilute one of them with water to scale and shake well. Use distilled water as a reference to determine the absorbance of the sample itself. The operation of the other one shall be carried out according to the relevant provisions of 4.1.4.1.2 to 4.1.4.1.3. The absorbance of glyphosate in the sample is obtained by deducting the absorbance of the sample itself from the measured absorbance. 4.1.4.3 Calculation of analytical results
The percentage of glyphosate (r1) is calculated according to formula (1): piVi
HG2162-91
Wherein: pr——mass concentration of glyphosate in the standard solution, mg/mL; Vi——volume of the standard solution corresponding to the absorbance of glyphosate in the sample on the standard curve, mL; P2——mass concentration of the sample solution, mg/mL; V2——volume of the sample solution absorbed, mL. 4.1.4.4 Allowable difference
The difference between the parallel determination results of this method shall not exceed 1%. Note: After the colorimetric blood is used up, it should be washed with nitric acid solution. 4.2 Determination of water-insoluble matter
4.2.1 Instrument
4.2.1.1 No. 3 glass sand core;
4.2.1.2 Vacuum pump;
4.2.1.3 Filter bottle: 1000mL.
4.2.2 Analysis steps
Weigh 10g of sample (accurate to 0.001g), place in a 500mL beaker, add 200mL of water to dissolve (slightly heat if necessary), transfer the solution to a No. 3 glass crucible with constant weight, filter with vacuum, drain and then wash three times with 100mL of warm water, drain. Bake the glass crucible at 1055℃ to constant weight (accurate to 0.0002g). 4.2.3 Calculation of analysis results
The percentage of water-insoluble matter (a2) is calculated according to formula (2): 22
Where: m1—the mass of water-insoluble matter when dried to constant weight, g; m2——the mass of the crucible when dried to constant weight, g;
m—the mass of the weighed sample, g.
4.3 Determination of pH valuebzxz.net
Determine according to the provisions of GB1601.
5 Inspection rules
5.150% glyphosate soluble powder should be inspected by the quality inspection department of the manufacturer. Water-insoluble matter is a type inspection item and should be inspected at least once a week. The manufacturer should ensure that all glyphosate soluble powders shipped meet the requirements of this standard, and each batch of products shipped should be accompanied by a quality certificate.
5.2 Users have the right to inspect whether the quality of the 50% glyphosate soluble powder received meets the requirements of this standard in accordance with the provisions of this standard.
5.3 The sampling method shall be carried out in accordance with the relevant provisions of GB1605. 5.4 When the sample is mixed, use the quartering method to reduce it and take out a sample of not less than 50g and put it into a clean and dry glass wide-mouth bottle. A label should be attached to the bottle, indicating the manufacturer's name, batch number, and sampling date, and it should be sent to the inspection department for inspection. 5.5 If one of the test results does not meet the requirements of this standard, it is necessary to re-sample from two times the packaging for re-testing. Even if only one of the test results does not meet the requirements of this standard, the entire batch of products cannot be accepted. 5.6 When the supply and demand parties have disputes over the product and need arbitration, the arbitration unit can be agreed upon by both parties. During arbitration, verification shall be carried out in accordance with the methods specified in this standard.
